چکیده

Abstract Background The identification of chromosomes among Avena species have been studied by C-banding and in situ hybridization. However, the complicated results from several cytogenetic nomenclatures for identifying oat are often contradictory. A universal karyotyping nomenclature system precise chromosome comparative evolutionary studies would be essential genus based on recently released genome sequences hexaploid diploid species. Results Tandem repetitive were predicted physically located chromosomal regions sativa OT3098 assembly v1. Eight new oligonucleotide (oligo) probes sequential fluorescence hybridization (FISH) designed then applied mitotic metaphase spreads A. brevis , nuda wiestii ventricosa fatua We established a high-resolution standard karyotype distinct FISH signals multiple oligo probes. painting with bulked oligos, wheat-barley collinear regions, was used to validate linkage group assignment individual chromosomes. integrated our Oligo-FISH earlier through methods, subsequently determined breakage points some translocations . Conclusions This will powerful tool describing genetic diversity, rearrangements relationships genomic approaches.

Discrimination of the closely related A and D genomes of the hexaploid oat Avena sativa L.

A satellite DNA sequence, As120a, specific to the A-genome chromosomes in the hexaploid oat, Avena sativa L., was isolated by subcloning a fragment with internal tandem repeats from a plasmid, pAs120, that had been obtained from an Avena strigosa (As genome) genomic library. Southern and in situ hybridization showed that sequences with homology to sequences within pAs120 were dispersed througho...

Comparative cytogenetic analysis of Avena macrostachya and diploid C-genome Avena species.

The chromosome set of Avena macrostachya Balansa ex Coss. et Durieu was analyzed using C-banding and fluorescence in situ hybridization with 5S and 18S-5.8S-26S rRNA gene probes, and the results were compared with the C-genome diploid Avena L. species. The location of major nucleolar organizer regions and 5S rDNA sites on different chromosomes confirmed the affiliation of A. macrostachya with t...

A molecular marker map in 'Kanota' x 'Ogle' hexaploid oat (Avena spp.) enhanced by additional markers and a robust framework.

Molecular mapping of cultivated oats was conducted to update the previous reference map constructed using a recombinant inbred (RI) population derived from Avena byzantina C. Koch cv. Kanota x Avena sativa L. cv. Ogle. In the current work, 607 new markers were scored, many on a larger set of RI lines (133 vs. 71) than previously reported. A robust, updated framework map was developed to resolve...

Transferability and utility of white oat (Avena sativa) microsatellite markers for genetic studies in black oat (Avena strigosa).

Preservation and use of wild oat species germplasm are essential for further improvement of cultivated oats. We analyzed the transferability and utility of cultivated (white) oat Avena sativa (AACCDD genome) microsatellite markers for genetic studies of black oat A. strigosa (A(s)A(s) genome) genotypes. The DNA of each black oat genotype was extracted from young leaves and amplified by PCR usin...
